Ammanford started slowly on the back of a narrow defeat to Caerbryn last week and conceded an early goal when keeper Darren Warnes was beaten after just ten minutes.
However, Kyle Wells and Matt Warnes soon took control of midfield for the hosts and began to play some neat passing football.
Ammanford battled hard and got their equaliser from a Joe Kinsey corner when captain Jack Conway slotted the ball home after a battling scramble in the Llandeilo penalty area.
The second half saw Ammanford assert their passing game and Jake Webb was able to score from a great long ball played by Dylan Thomas.
Webb got his second after a Lloyd Bevan free-kick was not cleared by Llandeilo and he fired home.
Warnes made it four with a cracking strike from long range.
Llandeilo got a goal back but it was too late and Ammanford can be pleased with their best display of the season against early promotion favourites Llandeilo.
Meanwhile, Llandeilo Firsts put eight past Johnstown to cruise into the next round of the Carmarthenshire League Challenge Cup.
